Metabolite quantitation has many advantages over untargeted non quantitative measurements. Better recovery by optimization of extraction, larger dynamic range by optimizing for sensitivity and better relative measurements by using linear calibration curves, along with better specificity. The degree of method validation for quantitative analysis depends upon use. For early discovery, validation can be streamlined, while for use in a clinical study (phase 2 or 3), validation packages can be more inclusive and designed by both client and HMT. Using capillary electrophoresis expands the metabolite range to very polar metabolites such as phosphate, pyrophosphate and inositol phosphates.
